Biobrick Vector choice

We have selected from BioBrick plate the pSB1A3 plasmid to clone 1Xore and 2Xore promoters and pho4 coding sequence. This is a high copy number plasmid (copy number of 100-300 per cell) carrying ampicillin resistance.

Instead we have selected J61002 and J61003 plasmids to clone respective Pho80 and Pho8 coding sequence. We decided to insert a promoter element between EcoRI and XbaI and Pho80 or Ph08 Cds with XbaI and SpeI.
